One of the most important events in the regulation of plant development is the transition from the vegetative to the reproductive phase. The precise control of this transition, which has a profound effect on grain production in annual temperate cereals such as wheat and barley, is determined mainly by seasonal changes in day length (photoperiod) and by winter-like temperatures (vernalization). Recent molecular and genetic analyses in wheat have revealed the molecular mechanisms underlying flowering responses of wheat to changes in photoperiod and cold temperature. Here, we describe genes related to vernalization, photoperiod, and earliness per se (eps), and the molecular mechanism regulating flowering time through vernalization and photoperiod genes in wheat.
서 언
식물의 생활사는 종자로부터 발아하여 유묘로 생장하는 영양 생장기와 꽃을 피워서 종자를 생성하는 생식 생장기로 크게 나누어진다. 특히, 영양 생장기에서 생식 생장기로 넘어가는 단계를 개화로 정의하며, 이 과정에 빛과 온도와 같은 다양한 외부 환경과 식물 자체의 발달 단계 및 식물 호르몬이 중요하게 영향을 주는 것으로 알려져 있다(
Simpson & Dean 2002). 식물이 성공적으로 개화를 유도하기 위해서 각 식물체는 개화시기를 정교하게 조절하기 위하여 다양한 유전적인 신호전달 네트워크를 진화시켜 왔다.
식물의 개화시기를 조절하는데 관여하며, 지금까지 300여개 이상의 유전자들이 서로 간에 복잡한 신호전달 네트워크를 형성하는 것으로 알려져 있다. 지난 30년간 모델 식물인 애기장대에서 다양한 돌연변이체들이 분리하고, 그 기능을 규명하여 적어도 5가지 중요한 개화시기 조절 경로가 있음이 밝혀졌다: 광주기 경로(photoperiod pathway), 춘화처리 경로(vernalization pathway), 자발적 경로(autonomous pathway), 주변온도 경로(ambient temperature pathway), 지베렐린 경로(gibberellin pathway) (
Boss et al. 2004,
Putterill et al. 2004,
Sung & Amasino 2004,
Lee et al. 2007,
Fornara et al. 2010,
Lee et al. 2013). 이러한 경로들을 통해서 받아들여진 개화를 유도하는 다양한 신호들은 ‘개화통합 유전자(floral integrators)’로 불리는 일부 유전자들의 발현을 조절하여 최종적으로 정단분열조직(shoot apical meristem)이 꽃을 형성할 수 있는 개화분열조직(floral meristem)으로 전환하게 한다(
Lee et al. 2006). 또한, 애기장대 연구를 토대로 해서 여러 채소작물, 벼, 나무 등에서도 개화시기를 조절하는데 관여하는 유전자들이 지속적으로 보고되고 있다(
Bohlenius et al. 2006,
Greenup et al. 2009,
Schiessl et al. 2017).
본 논문은 밀에서 개화를 조절하는 춘화처리(vernalization), 광주기(photoperiod), earliness per se (eps)에서 작용하는 loci를 중심으로 알아보고, 춘화처리와 광주기 유전자들이 작용하는 밀의 개화시기 조절 기작에 대해서 알아보고자 한다.
밀 개화시기의 유전학적 조절
밀의 개화 반응은 상당히 복잡하며 이와 관련된 유전자들에 다양한 변이가 존재하지만, 주로 춘화처리 유전자(
Vrn gene), 광주기 유전자(
Ppd gene),
earliness per se (
esp gene) 유전자들이 관여하는 것으로 알려져 있다(
Herndl et al. 2008,
Kamran et al. 2014). 따라서, 다양한 환경에서 경작되고 있는 밀의 환경에 대한 적응력과 생산성은 3가지 유전자들이 작용하는 개화시기 조절경로와 생육온도에 따른 이 조절 경로들간의 상호작용에 의해 결정되어진다(
Gororo et al. 2001). 일반적으로 적정 온도에서 밀의 개화를 위해서 광주기 및 춘화처리 유전자들은 다양한 환경변화에 따라 개화를 촉진하거나 지연시킨다(
Law & Worland 1997). 예를 들어, 춘화처리 유전자는 높은 북쪽 위도에서 생장하는 겨울밀의 개화를 지연시켜 극심한 추위로부터 밀의 생식 기관을 보호한다. 또한, 밀이 생장하기에 너무 더운 지역에서는 광주기 둔감 유전자가 늦겨울과 이른 봄에 밀의 개화를 촉진시킨다.
일반적으로 춘화처리 유전자, 광주기 유전자, 그리고
eps 유전자가 각각 70-75%, 20-25%, 5%가 밀의 개화와 관련된 유전적 다양성을 설명하는데 관여한다(
Stelmakh 1998). 밀의 개화에 영향을 주는 유전자들은 밀의 7개 염색체에 고루 분포되어 있고, 춘화처리, 광주기,
eps에 관여하는 유전자들도 각각의 3개의 상동 염색체에 위치하는 것으로 알려져 있다(
Law & Worland 1997).
춘화처리 유전자(vernalization gene)
밀의 생장 습성은 유전적으로 춘화처리에 대한 감수성을 결정하는 유전자들에 의해 조절되며, 여기에는
Vrn-1,
Vrn-2,
Vrn-3,
Vrn-4 유전자의 변이가 중요하게 영향을 미친다(
Flood & Halloran 1986,
Distelfeld et al. 2009a). 봄 밀과 조건적(facultative) 생장 습성을 보이는 밀에서는
Vrn-1,
Vrn-3,
Vrn-4 유전자들에 하나 이상의 우성 대립 형질이 발견이 되며, 이러한 유전적 변이는 춘화처리에 대해 반응을 하지 않도록 한다. 반면에 겨울 밀은 우성 대립형질의
Vrn-2 유전자와 열성 대립형질의
Vrn-1,
Vrn-3,
Vrn-4 유전자들을 가지고 있기 때문에 개화를 위해서는 춘화처리가 필수적이다.
Vrn-1 유전자들은 3개의 이종 상동성 유전자(orthologous gene)인
Vrn-A1,
Vrn-B1,
Vrn-D1을 각각 염색체 5A, 5B, 5D에 가지고 있다(
Pugsley 1971,
Dubcovsky et al. 1998,
Fu et al. 2005). 겨울 생장 습성을 조절하는데 관여하는
Vrn-2 유전자들은
Vrn-A2와
Vrn-B2가 2배체와 4배체 밀에서 특성이 규명이 되었으며,
Vrn-A2는
T. monococcum의 염색체 5A에 위치한다(
Yan et al. 2004,
Distelfeld et al. 2009b).
Vrn-3 유전자인
Vrn-A3,
Vrn-B3 (과거에는
Vrn-5나
Vrn-D4로 명명),
Vrn-D3는 6배체 밀에서 각각 염색체 7A, 7B, 5D에 위치한다(
Yan et al. 2006,
Bonnin et al. 2008,
Yoshida et al. 2010).
Vrn-3 유전자들은
Vrn-1 유전자들의 전사를 촉진시켜 밀의 조기 개화를 유도하는 것으로 알려져 있다(
Yan et al. 2006). 앞서 언급한 3개의
Vrn (
Iwaki et al. 2000) 유전자들 외에
Vrn-4 유전자에 대한 대립형질인
Vrn-D4가 염색체 5D에 위치하는 것으로 알려져있지만, 이 유전자의 기능에 대해서는 아직까지 규명이 되지 않았다(
Iwaki et al. 2000). 그러나, 최근 연구에 따르면
Vrn-D4 유전자가
Vrn-1,
Vrn-2,
Vrn-3 유전자들의 상위 혹은
Vrn-1,
Vrn-2,
Vrn-3 유전자들이 관여하는 feedback regulatory loop과 연관될 가능성이 보고가 되었다(
Kippes et al. 2014).
광주기 유전자(photoperiod gene)
광주기 반응은 밀의 개화시기를 결정하는데 있어서 두 번째로 중요한 유전적 시스템으로 밀이 서로 다른 기후 조건에 적응하여 생장하도록 한다. 광주기는 주로 다른
Vrn 유전자들에 영향을 받지 않는 가을에 씨앗을 파종하는 봄 밀이나 겨울 밀 품종의 개화에 영향을 주는 것으로 알려져 있으며, 이러한 밀 품종들에서 광주기에 대한 감수성이 중요하게 개화시기를 결정한다(
Worland et al. 1994,
Worland et al. 1998,
Kamran et al. 2014). 광주기 둔감 품종은 봄에 온도가 올라가게 되면 바로 영양 생장기에서 생식 생장기로 전환이 되지만, 광 민감 품종은 낮 길이가 길어져서 광주기 충분 요건을 갖추어질 때에만 개화를 하게 된다.
광주기에 대한 반응을 결정하는 유전자들은
Ppd 유전자로서 밀의 염색체 2번에 위치하는 것으로 알려져 있다(
Worland et al. 1994,
Worland et al. 1998). 지금까지
Ppd-A1,
Ppd-B1,
Ppd-D1 유전자들(과거 논문에서는 각 유전자들이
Ppd3,
Ppd2,
Ppd1으로 보고가 됨)이 밝혀져 있고, 이들은 각각 염색체 2A, 2B, 2D에 위치하고 있다(
Law et al. 1978,
Scarth & Law 1983).
Vrn 유전자들과 유사하게 우성 대립형질인
Ppd 유전자는 낮 길이에 대해 둔감성을 나타내게 하고, 열성 대립형질인
Ppd 유전자는 낮 길이에 대해 민감성을 나타내게 한다(
Pugsely 1966).
Ppd-A1,
Ppd-B1,
Ppd-D1 유전자들 외에 밝혀진 광주기 반응 유전자는
Ppd-B2가 있으며, 밀의 염색체 7BS에 위치하고 있다(
Khlestkina et al. 2009). 이 유전자는 낮 길이에 관계없이 개화를 촉진시키는 잘 알려진
Ppd 유전자와 달리 오직 장일 조건에서만 개화를 촉진한다. 특히, 유전학적 분석을 통해서
Ppd-D1 유전자가
Ppd-B1,
Ppd-A1 유전자들에 비해 가장 효과적인 광주기 둔감 유전자로 밝혀졌으며,
Ppd-D1과
Ppd-A1 유전자의 대립형질의 변이가 광주기 반응에 따른 밀의 개화시기 결정에 중요하다(
Worland 1996,
Whittal et al. 2018). 밀의 염색체 2번에 위치하는
Ppd-A1,
Ppd-B1,
Ppd-D1 유전자들 외에 다른 염색체에도 밀의 개화시기에 영향을 미치는 광주기 반응 유전자들이 존재한다. 예를 들어, 염색체 1번에는 춘화처리와 광주기 모두에 반응하여 개화시기를 지연시키는 유전자가 존재하며, 염색체 3D에는 광주기 둔감 유전자가 위치한다(
Miura & Worland 1994,
Law et al. 1998).
Earliness per se 유전자(eps gene)
광주기 및 춘화처리 반응 외에도 밀의 개화시기에 영향을 주는 유전적 요인이 알려져 있으며, 이러한 유전적 차이를 “earliness per se”라고 부른다(
Halloran 1975,
Ford et al. 1981,
Scarth & Law 1983,
Hoogendoorn 1985,
Kamran et al. 2014).
eps 유전자는 춘화처리와 광주기의 필요성이 충족이 된 밀 품종의 개화시기 차이에 관여하며, 환경 변화에 관계없이 개화시기에 영향을 준다(
Worland 1996,
Kato et al. 1998). 주요 춘화처리 및 광주기 유전자들은 특정 환경 조건에서만 반응을 해서 개화시기에 영향을 주기 때문에 “earliness의 조절인자”로 알려져 있지만, earliness는 외부 환경에 반응하여 독립적으로 화아(floral primordia) 분화를 개시할 수 있는 최소 생식 생장에 영향을 준다(
Kato & Wada 1999). 춘화처리 및 광주기 반응은 개화시기에 주요 영향을 미치기 때문에 자세하게 연구가 되어 왔지만,
eps 유전자는 많이 밝혀져 있지 않았다.
eps 유전자는 춘화처리 및 광주기 유전자의 효과가 없을 시에만 개화시기에 영향을 주며, 일부 유전자들이 관여하는 양적 형질로 알려져 있다. 그러나,
eps 유전자는 개화시기에 영향을 주는 환경 요인에 독립적으로 밀의 생활사를 단축시키기 위한 육종 프로그램에 효과적으로 이용될 수 있다.
밀의 유전 집단을 대상으로 한 개화시기에 대한 유전적 분석을 통해서
eps 유전자들은 상대적으로 개화시기에 작은 영향을 미치며, 일반적으로 양적 형질로 작용하는 것으로 알려져 있다(
Kato et al. 1999a,
1999b). 춘화처리 및 광주기 유전자들의 효과가 없는 환경 조건과 특정 밀 품종을 이용한 유전적 분석은
eps 유전자들이 밀의 다른 중요한 농업 형질을 제외한 개화시기에만 영향을 미치는 것으로 알려져 있다.
eps 유전자 및 이와 관련된 양적 형질들은 밀의 모든 염색체에 위치하는 것으로 보고가 되었지만, 주로 염색체 5A, 2B, 3A, 5B, 7B, 그리고 4D에 주로 위치하고 있다(
Kamran et al. 2014).
밀의 개화시기 조절 기작
밀이 넓은 환경 조건에 적응해서 생육할 수 있는 것은 생육 조건을 조절하는 춘화처리 유전자들과 광주기 유전자들의 대립형질의 다양성에 의해 결정된다. 광주기 유전자들의 차이는 광 민감 밀과 광 둔감 밀로 구분하고, 춘화처리 유전자들의 차이는 겨울 밀과 봄 밀로 구분한다. 겨울 밀은 가을에 심어 개화를 촉진시키기 위해 긴 기간의 저온 상태(춘화처리)가 필요하고, 봄 밀은 일반적으로 봄에 심어 춘화처리를 필요로 하지 않는다(
Distelfeld et al. 2009a).
춘화처리 요구에 필요한 자연변이(natural variation)는 춘화처리 유전자인
VRN1,
VRN2,
VRN3의 대립형질의 차이와 밀접한 관련이 있다(
Distelfeld et al. 2009a).
VRN1 유전자는 애기장대 분열조직 정체성 결정 유전자인
APETALA1 (
AP1),
CAULIFLOWER (
CAL),
FRUITFULL (
FUL)과 높은 상동성을 가진 MADS-box 전사조절인자를 암호화하며, 영양 정단분열조직을 생식 정단분열조직으로 전환하는 것을 조절한다(
Danyluk et al. 2003,
Trevaskis et al. 2003,
Yan et al. 2003).
VRN1 유전자의 결실 돌연변이를 가진 밀은 개화를 하지 못하기 때문에 개화에 중요한 역할을 하는 것으로 알려져 있다. 최근 연구에 따르면
VRN1 유전자의 발현은 탄수화물 결합 단백질을 암호화하는 유전자인
VERNALIZATION-RELATED 2 (
VER2)와 RNA 결합 단백질인 TaGRP2의 상호작용에 의해 조절이 된다고 알려져 있다(
Xiao et al. 2014).
VRN2 유전자는 애기장대 유전자와 상동성이 없는 하나의 zinc finger와 CCT 도메인을 가진 2개의 ZCCT 유전자들이며, 이 유전자들의 결실을 가진 밀은 봄 밀의 생육 성질을 가지기 때문에 개화 억제자로 작용하는 것으로 알려져 있다(
Yan et al. 2004,
Distelfeld et al. 2009b).
VRN3 유전자는 애기장대 FLOWERING LOCUS T (FT) 단백질과 높은 상동성을 가진 RAF kinase inhibitor 단백질을 암호화하며, 개화를 촉진하는 것으로 알려져 있다(
Yan et al. 2006,
Li & Dubcovsky 2008). 광주기 반응에 대한 자연변이(natural variation)는
PSEUDORESPONSE REGULATOR (
PRR) 유전자 군의 member인
PPD1 유전자의 대립형질의 차이에 의해 주로 결정되며, 광 민감 밀에서
PPD1 유전자는 장일 조건에서
VRN3 유전자의 발현을 증가시켜 개화를 촉진한다(
Turner et al. 2005). 이러한 유전자들 외에도 애기장대 FT 단백질과 결합을 하는 bZIP 전사조절인자인 FD 단백질과 상동성을 보이는 FD-LIKE2 (FDL2) 단백질과 애기장대 CONSTANS (CO) 단백질의 CCT 도메인과 결합을 하는 HEME ACTIVATOR PROTEIN (HAP) 단백질이 밀에서 밝혀졌다(
Li & Dubcovsky 2008,
Distelfeld et al. 2009b).
밀의 개화시기는 개화에 직접적인 영향을 주는
VRN1 유전자의 발현에 영향을 주는 춘화처리 및 광주기와 같은 환경에 의해 매개되는 개화 억제 인자와 촉진 인자간의 균형에 의해 조절된다(
Distelfeld et al. 2009a,
Chen & Dubcovsky 2012,
Fig. 1).
VRN3 유전자는 광주기 유전자들인 PPD1-CO module과 춘화처리 유전자들인 VRN1-VRN2 module에 의해 매개되는 환경 신호를 통합한다. 이 과정에서 CCT 도메인을 가진 CO 단백질과 VRN2 단백질은 trimeric complex인 HAP2/3/5의 CCT 도메인과 상호작용을 하여 장일 조건에서 PPD1-CO module과 VRN1-VRN2 module에 오는 개화의 신호를 조절하게 된다. VRN3 단백질과 FDL2 단백질간의 결합은
VRN1 유전자의 발현을 증가시킨다. 춘화처리는 장일과 단일 조건에서
VRN1 유전자의 발현을 증가시켜
VRN2 유전자의 발현을 억제한다. 또한,
VRN1 유전자의 발현은 VER2 단백질과 TaGRP2 단백질의 상호작용에 의해 조절이 된다: 춘화처리는
VER2 유전자의 발현과 TaGRP2 단백질의
O-GlcNAcylation 수준을 증가시키며, 이를 통해 인산화된 VER2 단백질이 핵으로 이동하여
O-GlcNAcylated TaGRP2 (TaGRP2-G) 단백질과 결합하여 TaGRP2 단백질에 의한
VRN1 유전자의 발현 억제를 풀리게 한다(
Xiao et al. 2014). 또한 밀의 개화시기 조절에서 3개의 춘화처리 유전자들인
VRN1,
VRN2,
VRN3간의 상호작용이 feedback regulatory loop을 구성하며, 이를 통해 개화가 개시가 된다.

전 망
International Wheat Genome Sequencing Consortium (IWGSC)는 2005년부터 전게놈 염기서열 분석을 시작하여 13년 만에 2018년 8월에 6배체 밀 21개 염색체 염기서열 정보(Chinese Spring, 약 98%, 13.8 Gb, 107,891개 유전자)를 완전히 제공하였다(
International Wheat Genome Sequencing et al. 2018). 또한, GMO (genetically modified organism)의 사회적 문제로 인하여 식물생명공학자들에게 유전자편집기술 중 하나인 CRISPR/Cas9 시스템을 통한 식물 유전체의 변형이 중요한 기술로 부각되고 있다(
Scheben et al. 2017). 하나의 실제 적용 사례로 애기장대
FLOWERING LOCUS C (
FLC)의 밀의 상동 유전자인 3개의
TaAGL33A,
TaAGL33B,
TaAGL33D에 대한 유전자에 대한 편집이 성공적으로 수행이 되어 6배체 밀의 개화가 앞당겨졌음이 보고가 되었다(
International Wheat Genome Sequencing et al. 2018). 국내에서는 국내 밀 자원에 대한
Vrn-1 및
Ppd-1 loci에 대한 유전적 조성 및 숙기 관련 특성과 수량 관련 표지인자 탐색에 관련된 연구가 이루어졌지만, 제한된 재료에 대한 평가로 인해 밀 숙기 및 수량성 증진을 위한 육종 프로그램의 적용에는 한계가 있다(
Lee et al. 2014,
Cho et al. 2015a,
Cho et al. 2015b,
Cho et al. 2018). 따라서, 최근에 밝혀진 6배체 밀의 염기서열정보를 통해 향후에 한국형 밀에 대한 표준 염기서열정보를 구축하고, 유전자 편집 기술을 이용한 밀 개화시기 유전자들에 대한 “역유전학적(reverse genetics)” 분석을 통해 해당 유전자들의 개화시기 조절에서의 기능이 구체적으로 밝혀질 것이다. 또한, 한국형 밀을 대상으로 한 춘화처리, 광주기,
eps 유전자들에 대한 발굴과 이 유전자들간의 상호작용을 통한 밀 개화시기 조절 기작에 대한 분자적 수준에서의 연구도 더불어 수행이 되어야 할 것이다.
적 요
식물의 생장을 조절하는 가장 중요한 과정 중의 하나는 영양 생장기에서 생식 생장기로 전환되는 것이다. 이러한 생장기의 전환은 낮의 길이(광주기)와 겨울과 같은 저온(춘화처리)의 계절적 변화에 의해 주로 정확하게 조절이 되며, 이는 밀과 보리와 같은 일년생 곡물의 생산량에 중요한 영향을 미친다. 최근의 밀을 이용한 분자 유전학적 분석들은 광주기와 저온에 반응하는 유전자들을 통한 밀의 개화 반응에 관련된 분자 생물학적 조절 기작을 규명하였다. 이 리뷰에서는 밝혀진 밀의 춘화처리, 광주기, eps 유전자들 및 춘화처리와 광주기 유전자들을 통한 밀의 개화시기 조절 기작에 대해 기술하였다.
