Artificial selection of ginseng has been practiced since Hwangsook (with yellow pericarp and a green stalk, and was developed from a landrace parent) and Cheonggyeong (with red pericarp) were selected as breeding lines in 1926. Systematic research into ginseng breeding, however, started in earnest in the 1960s when the Central Research Institute of Monopoly and Technology (CRIMT) was established, and the Korean Ginseng Experiment Station was organized under the CRIMT. Research into variant characteristics, resource collections, and genetic evaluations began around this time. With the establishment of the Korean Ginseng Institute in the 1970s, studies involving pedigree selection, cataloguing of agricultural traits of genetic resources, generation shortening by tissue culture, and heritability assessments were conducted. In the 1980s, regional adaptation tests were carried out on breeding lines, focusing on ginseng-producing districts. In the 1990s, research was performed on seed multiplication for variety diffusion, effective components and processing quality, and cross breeding. Foreign ginsengs were introduced for interspecies hybridization, and studies were conducted using genetic engineering techniques. Since the 2000s, applications have been made to patent different ginseng cultivars. Currently, 32 cultivars are registered at the Korea Seed & Variety Service. Future goals for ginseng breeding include developing climate change- and disaster-resistant, consumer-oriented, high-performance cultivars. Therefore, it is necessary to develop technologies for distributing new cultivars by collecting and evaluating genetic resources, and cross breeding and performing mass propagation using these resources.

최근 전 세계인의 웰빙과 건강에 관한 관심 증가로 다양한 한약재를 원재료로 한 건강기능식품이 등장하고 있다. 인삼은 이러한 다양한 한약재 중에서도 대표적인 한약재로 손꼽히며 전 세계적으로 널리 이용되고 있다. 특히 최근에는 베트남 등의 동남아시아와 유럽 등에서 인삼에 대한 관심이 높아지고 있다.
고려인삼(
Panax ginseng C. A. Meyer)은 우리나라의 중요한 천연자원으로서 예로부터 영약(靈藥) 또는 선약(仙藥)으로 사용됐다. 중국의 문헌에 처음으로 기재되어 있는 것은 사유의 급취장(전한 원제시대, B. C. 33~48) 이며, 장중경의 상한론(후한 헌제시대, B. C. 196~200)에는 총 113개 처방 중 21개의 인삼 배합 처방이 수록되어 있다. 그 후 신농본초경, 신농본초경집주, 명의별록, 방약합편, 경악전서, 동의보감 등 많은 의서에 보기 약재로의 인삼 기록이 수록되어 있다(
Lee 2013).
Panax 속에 속하는 식물은
Panax ginseng C. A. Meyer를 비롯하여
P. quinquefolius,
P. notoginseng,
P. japonicus,
P. trifolius 등 17종이 알려져 있다. 이 중에서 Asian ginseng으로 알려진
P. ginseng (한국, 중국, 일본 등 동아시아 지역 재배 기원), American ginseng으로 알려진
P. quinquefolium (캐나다와 미국 동부 및 중국 재배 기원) 및 삼칠삼으로 불리는
P. notoginseng (중국 남서부 및 베트남 재배 기원)의 3종이 주로 재배되어 한약재로 널리 유통되고 있다(
Wen & Zimmer 1996).
고려인삼은 아시아의 극동지방 북위 34~48° 사이의 우리나라 남해 지역과 제주도를 제외한 지역(34~43°), 중국 동북지역(43~47°) 및 러시아 지역(42.5~48°)에서 자생하고 있으며, 상당히 제한적인 범위의 위도와 경도 상에서 생육할 수 있다(
Woo et al. 2004). 우리나라에서 생산된 인삼이 다른 나라에서 생산되는 삼보다 높이 평가받는 이유는 재배 적지가 좁음에도 불구하고 계절의 변화가 다른 지역보다 분명하고 생육 기간이 긴 특성이 있기 때문이다(
Kim 2012).
고려인삼의 주요 약리 물질들은 사포닌(진세노사이드류), 페놀 화합물, 다당류, 펩타이드류, 폴리아세틸렌류, 지방산 등으로 알려졌지만(
Attele et al. 1999), 이 중에서 사포닌 성분이 가장 대표적인 약리효능 물질로서 주목을 받고 있다. 사포닌은 비누와 같은 특징을 갖는 배당체(glycosides)로써, 수많은 식물 종에서 생성된다. 또한 수백 년 전부터 약리적 활성을 인정받았으며 비누풀(
Saponaria officinalis L.)에서 추출하여 비누로 사용하기도 하였다(
Osbourn 1996).
한편 인삼 종의 기원을 동정하기 위해서 사포닌이 활용되고 있으며, protopanaxadiol 계와 protopanaxatriol 계의 사포닌이 다양한 약리 작용을 가지고 있기 때문에 품질 관리의 지표 성분으로 이용되고 있다. 또한 사포닌 종류에 따라 다양한 질병에 대한 작용도 다른 것으로 알려져 있다(
Elma et al. 1991,
Henishin et al. 1991,
Park et al. 1991). 사포닌의 함량은 주로 온도, 습도 등 재배 환경에 따라 차이가 나지만 품종이나 계통에 따라서도 차이가 있을 것으로 추측되어 앞으로 성분(기능성) 육종에 대한 연구가 수행되어야 할 것으로 판단된다.
앞서 기술한 바와 같이 인삼을 대상으로 한 연구는 다른 식량 작물에 비교하였을 때, 주로 생리 활성, 효능 등에 관련된 연구가 진행되어 왔으며, 최근에 와서야 분자 수준에서의 유전학과 생리학 등의 기초과학 분야에 관련된 연구가 시작되고 있다. 한편 인삼 품종의 역사는 다른 작물에 비해 짧은 편이어서 앞으로도 많은 연구가 진행되어야 할 것으로 판단된다.
우리나라 인삼 육종의 주요성과
고려인삼은 산형목 오가과(
Araliaceae)
Panax속 다년생 식물이다. 지역적으로 차이가 있으나 출아는 매년 4월 중순경에, 개화는 5월 초순경에 이뤄진다(
Kwon et al. 2001). 꽃은 5장의 꽃잎과 꽃받침에 1개의 암술과 5개의 수술 및 2개의 주두를 갖는 완전화이다. 수정은 자가수정을 원칙으로 하지만 간혹 충매, 풍매 등에 의해 타가 수정되기도 한다(
KT&G 1996). 꽃은 꽃대의 꼭대기 끝에 여러 개의 작은 꽃(소화, 小花)들이 방사형으로 달려 하나의 큰 꽃송이처럼 보이는 산형화서를 이루고 있는데, 산형화서의 가장자리 소화(小花)들부터 개화하기 시작하여 점차 꼭대기 중심 부위로 개화가 진행되는데, 완전히 개화되는데 소요되는 시간은 7~15일이다. 또한 1개의 화경에 40여 개의 열매가 맺히는 핵과류이다(
Rhim 2010). 종자는 대체로 1과에 2립이 보통이지만 드물게는 1과에 1립 또는 1과에 3립인 경우도 있다(
KT&G 1996).
현재까지 국내에서 인삼 품종 육성은 제한된 수의 품종만이 개발되어 왔으며, 이러한 품종들을 지역 재래종에 비교하여 뿌리의 형태가 양호하고, 병해충에 강하며, 사포닌을 다량으로 함유하고 있고, 수량성이 좋은 고유한 형태적 특성과 더욱 나은 농업적 특성을 보이고 있다(
Kim et al. 2013,
Kwon et al. 2003).
인삼은 종자를 파종하여 다시 종자를 얻기까지 최소 3~4년이 소요되며, 인삼 지상부의 전형적인 특성을 파악하기 위해서 4년생부터 가능하다. 인삼은 주로 순계분리 육종법에 의해 육성을 하고 있는데, 이 방법을 이용할 경우 선발, 증식, 특성평가, 생산력 검정 시험, 지역 적응시험을 거치면 대략 36년이란 시간이 소요된다. 또한 교잡육종법을 이용하여 새로운 품종을 만들 경우 잡종 1대를 육성하고 분리 세대(F
2~F
5) 및 F
6 세대 이후의 선발 등의 단계를 거치면 산술적으로 60년 이상의 오랜 시간이 소요된다(
Cho 1998). 현재까지 육성된 품종은 모두 순계분리육종법에 의해 육성된 것으로 교잡육종법에 의해 육성된 품종은 아직 없는 실정이다.
국내에서 인삼의 육종이 시작된 시기는 1920년대이다. 1926년에는 경기도 개풍군에서 장과의 색깔이 황색을 띠는 황숙종이, 경기도 장단군에서 줄기의 색깔이 녹색인 청경종이, 경기도 개풍군에서 열매 색이 자경종과 황숙종의 중간인 등황색을 나타내는 등황숙종이 발견되었다. 국내 인삼 재배농가에서 이러한 변종들이 발견되면서 인삼의 유전자원 선발이 시작되었다(
Chung et al. 1992).
인삼 신품종 육성 분야에서 체계적인 연구가 시작된 시기는 1962년 중앙전매기술연구소 산하에 고려인삼시험장이 설치된 이후이다. 한편 농가에서 발견된 자경종과 황숙종을 대상으로 특성 비교 연구는 1963년에 수행되었고, 유전자원 수집 및 특성 검정은 1966년에 시작되었다(
KT&G 1972). 한편 1964년에 일본 나가노 시험장 Miyazawa에 의해서 미마끼(Mimaki)가 품종 등록되어 1967년에 우리나라에 도입되었으나, 품종의 구별성이 없어 품종으로 지속을 못 하고 있으며 현재 육종과 재배 연구가 중단되고 있다(
KT&G 1992).
1970년대에는 고려인삼연구소가 설립되었으며, 1980년대는 고려인삼연구소가 한국인삼연초연구소로 확대 개편되면서 인삼 품종 육성에 관한 연구 영역도 확대되었는데 농가에서 수집되어 선발 육성된 계통들을 대상으로 지역 적응시험이 본격적으로 수행되었다(
Kwon et al. 1991).
1990년대에는 순계분리 육종과 함께 교배육종도 병행되었는데 종간 교잡을 목적으로 미국삼(
P. quinquefolius L.), 삼칠삼(
P. notoginseng Burkill), 죽절삼(
P. japonicus C. A. Meyer) 및 히말라야삼(
P. pseudoginseng Wall)이 국내에 도입되었다(
Cheon et al. 1985). 인삼 종간 교잡의 경우 고려인삼과 미국삼을 대상으로 연구가 시도되었으나 F
2 세대에서 종자가 맺히지 않는 것으로 알려져 있다.
본격적으로 인삼 품종이 등록된 시기는 2000년대 이후부터이다. 한국인삼공사에서는 2002년에 천풍과 연풍을, 2003년에 고풍, 선풍, 금풍을, 2006년에 선운, 선원을, 2007년에 청선을 그리고 2009년에 선향을 품종 등록하였다. 2000년 초반에는 한국인삼공사가 민영화되면서 한국인삼공사에서 보존 중인 유전자원 317점이 농촌진흥청으로 이관되어 국가기관에서 인삼 육종을 담당하게 되었다.
또한 2000년대에는 국가기관인 농촌진흥청을 비롯하여 지역 특화작목연구소에서도 본격적으로 인삼 육종 연구가 시작되었다. 경기도원에서는 2012년에 K-1을, 충남도원에서는 2013년에 금선을, 2015년에 금진을, 농촌진흥청에서는 2013년에 천량을, 2015년에 고원을, 2019년에 천명과 진원을 품종 등록하였다(
Kim 2012,
Kim et al. 2013,
Kim et al. 2017). 이들 품종은 다수성이면서 병(점무늬병)과 고온에 강하며 적변(뿌리가 빨갛게 변하는 현상) 등이 없는 특성을 나타내고 있다(
Fig. 1).
현재까지 인삼 품종은 약 15%가 보급되었으며 아직도 85%는 지역 재래종에 의존하고 있다. 따라서 우리가 육성한 신품종이 보다 빨리, 더욱 많이 보급되기 위해서는 농촌진흥청과 지역 특화작목연구소(금산인삼약초시험장, 풍기인삼연구소 등) 등에서 개발된 품종을 현장실증사업과 신기술보급사업 등을 통하여 선도 농가를 중심으로 보급을 확산 시켜 나아가야 할 것이다.
한편 인삼 육종의 생명공학기술 적용은 2000년대 초반부터 활성화 되었으며 주로 인삼염기서열의 해독과 변이 탐색에 초점이 맞춰져 연구가 진행되었다. 게놈사이즈를 측정하기 위한 박테리아성 인공 염색체(Bacterial artificial chromosome, BAC) library 구축(Hong et al. 2004)과 상보적인 DNA 정보를 획득하기 위한 발현유전자 단편(expressed sequence tag, EST) 분석 연구 등이 이루어졌다(
Kim et al. 2006,
Sathiyamoorthy et al. 2010,
Choi et al. 2013).
결 론
인삼 품종 육성은 한국과 중국을 중심으로 경쟁 우위의 품종을 만들기 위해서 눈에 보이지 않는 치열한 경쟁을 벌이고 있다. 하지만 주로 생산자 중심의 품종(내재해성, 내병성) 육성이 이루어져 소비자와 산업체가 요구하는 품종(기능성) 개발도 필요한 실정이다. 현재까지 한국은 32개 품종이, 중국은 13개 품종이, 일본은 2개 품종이 육성되었는데, 이중 소비자 중심의 품종은 약 10% 정도에 불과하다. 따라서 인삼의 유효성분(사포닌계, 비사포닌계 등)이 많이 함유된 기능성 품종 육성이 필요하다.
인삼 가공 제품 수출은 대부분 홍삼이 차지하고 있고 중국 등 아시아권 수출 위주로 시장이 형성되어 있다. 수출 확대를 위해서는 수입국 맞춤형 품종 육성과 제품 개발을 통해 제품의 다양화와 중저가, 저가 등 다양한 해외시장 확대가 요구된다.
인삼 신품종은 2000년대 초반 한국인삼공사를 중심으로 육성되었으며, 최근에는 농촌진흥청과 지방 특화작목연구소 등 농촌진흥기관을 중심으로 품종이 육성되고 있다. 하지만 인삼의 신품종 보급률은 2018년 기준으로 14.6%로 낮은 실정이고 일부 품종(연풍 7%, 금풍 5%, 천풍 2%)만 보급되고 있다.
인삼은 종자 증식 배율이 매우 낮아 증식에 오랜 기간이 소요된다. 하나의 품종이 만들어져 보급에 필요한 양을 확보하기까지는 약 12년이 소요된다. 따라서 종자 보급 기간 단축을 위한 신품종 대량증식 기술과 종자 채종 효율성 향상 기술 개발이 필요하다. 또한 인삼 종자를 안정적으로 보급할 수 있는 신품종 보급체계 구축도 필요하다.
인삼은 유전자원의 다양성이 작고 한 세대가 4년으로 세대 고정에 오랜 기간이 소요되는 등 육종효율이 낮은 작물로 알려져 있다. 최근에는 인삼 육종효율을 높이기 위해서 다양한 생명공학기술이 접목되고 있다. 고온과 염류와 관련된 바이오마커를 개발하여 품종 육성에 활용하고 있으며, 조직배양을 이용하여 신품종을 대량증식하는 연구 등이 진행되고 있다.
인삼 신품종 육성과 보급을 위해서는 우선 이상기상에 대응하고 다양한 트렌드를 반영한 품종 육성에 집중하여야 할 것이다. 이를 위해서는 생산자 중심의 고온과 염류 등에 강한 품종과 병해에 강한 품종을 육성하고, 산업체와 소비자의 요구를 반영한 가공적성과 기능성이 우수한 품종을 육성해야 한다. 또한 선도 농가를 중심으로 채종포를 운영하여 신품종 보급을 확대해야 한다. 이를 위해서는 현장실증연구와 신기술보급사업 등을 활용하여 신품종 보급에 필요한 종자량을 먼저 확보해야 할 것이다. 마지막으로 바이오마커, 조직배양 등의 생명공학기술을 보다 적극적으로 활용하여 인삼 육종 효율성 향상을 위해서도 노력을 해야 할 것이다.
적 요
인삼은 1920년대 황숙종과 청경종이 발견되면서 유전자원 선발이 시작되었는데, 이때를 인삼 육종의 시작으로 본다. 그러나 인삼 육종에 대한 체계적인 연구는 1960년대 중앙전매기술연구소가 설립되고 산하에 고려인삼시험장이 설치되면서 본격적으로 이뤄지게 되는데, 이때 변종 간의 특성 연구, 자원 수집 및 평가 등이 시작되었다. 1970년대에 고려인삼연구소가 설립되면서 인삼 육종은 본격적으로 실시되는데, 이 시기에는 선발 계통의 체계적인 조사, 조직배양에 의한 세대 단축, 유전력 검정 등의 연구가 이뤄졌고, 이후 1980년대에 산지를 중심으로 우량 계통 지역적응시험이 실시되었다. 1990년대에 이르러 품종 보급을 위한 종자 증식과 함께 성분과 가공 품질에 대한 연구, 교배육종, 종간 육종을 위한 외국삼 도입, 유전공학 기법을 이용한 연구 등이 진행되었다. 2000년대에 이르러서는 본격적으로 품종이 등록되기 시작하여 현재 32개 품종이 등록되었다. 앞으로의 인삼 육종 방향은 기후변화에 대응한 병 및 재해 저항성, 소비자 맞춤형 고기능성 품종 등을 들 수 있다. 이를 위해서는 다양한 유전자원의 수집과 평가, 이들 자원을 활용한 교배와 대량증식 기술 개발을 통한 신품종 보급기술 개발 등이 필요할 것으로 생각된다.
