Integrating RNA Sequencing and Quantitative Trait Locus Mapping to Identify Potential Candidate Genes for Flooding Tolerance in Soybean

Flooding stress causes a significant reduction in soybean yield. The development of flood-tolerant cultivars is an effective way to minimize yield loss due to flooding. Information on candidate genes for flooding tolerance is useful for developing tolerant lines. The objective of this study was to identify potential candidate genes for flooding tolerance in soybean by integrating the results of a quantitative trait locus analysis and RNA sequencing. A total of 19 genes showed good amplification in capillary electrophoresis and were further analyzed through a reverse transcription quantitative polymerase chain reaction (qRT-PCR); two of these genes showed differential expression among tolerant and susceptible lines. The expression of Glyma.12g030900 and Glyma.10g050300 in leaf and root tissues, respectively, was higher in several tolerant lines than in the susceptible lines under flooding stress. The chlorophyll index of the tolerant lines was also consistently higher than that of the susceptible lines over two years, supporting the qRT-PCR results. This study provides useful information on flooding tolerance in soybeans.

A Virginia Typed Short Stem and Large Grain Peanut Variety ‘Ahwon’

A new peanut variety ‘Ahwon’(Arachis hypogaea ssp. hypogaea L.) was developed at the Department of Southern Area Crop Science, National Institute of Crop Science (NICS) in Milyang 2015. This was developed from the cross between ‘YG75’ with Shinpung-typed large grain and ‘YG46’ with Virginia-typed short stem. ‘Ahwon’ which is a Virginia plant type had 20 branches per plant and its length of main stem was 40 cm. Each pod has two grains with brown testa and long ellipse-shaped large kernel. Its yield components showed 40 pods per plant, 108 g of 100-seed-weight and 77% of pod shelling ratio in the regional yield trials (RYT). Seed quality showed 47.7% of crude oil and 29.4% of protein content. This variety showed resistant to early leaf spot and had more resistant to web blotch, stem rot and lodging, compared to reference variety. In the regional yield trials for 3 years ‘Ahwon’ was more productive than reference variety by 10% with 4.74 MT/ha for grain production.

A Shinpung Typed Large Grain and High-Yielding Peanut Variety ‘Daan’

A new peanut variety ‘Daan’(Arachis hypogaea ssp. hypogaea L.) was developed at the Department of Southern Area Crop Science, National Institute of Crop Science (NICS) in Milyang 2014. This was developed from the cross between ‘Iksan 31’ with Virginia typed short stem and ‘Dakwang’ with Shinpung-typed larger grain. ‘Daan’ which is a Shinpung plant type had 13 branches per plant and its length of main stem was 44cm. Each pod has two grains with brown testa and long ellipse-shaped large kernel. Its yield components showed 34 pods per plant, 127 g of 100-seed-weight and 75% of pod shelling ratio in the regional yield trials (RYT). Seed quality showed 47.8% of crude oil and 28.3% of protein content. This variety showed resistant to early leaf spot and had more resistant to late leaf spot, stem rot and lodging, compared to reference variety. In the regional yield trials for 3 years ‘Daan’ was more productive than reference variety by 16% with 5.00 MT/ha for grain production.

A new soybean variety, ‘Joongmo 3009’ (Milyang 222) was developed at the National Institute of Crop Science (NICS) in 2012. ‘Joongmo 3009’ was released by pedigree selection from the cross between ‘Cheongja 2(Milyang 121)’ and ‘Daemangkong’.

It has determinate growth habit, white flower, brown pubescence, brown pod color, green seed coat, green cotyledon, spherical seed shape, oval leaf shape and large seed size (29.3 grams per 100 seeds). It was late 16 days in maturing date than the check cultivar ‘Cheongjakong’. The average yield of ‘Joongmo 3009’ was 2.91 ton per hectare, which was higher 36 percentage than the check variety, in the regional yield trials carried out in three adaptable locations of Korea from 2010 to 2012. The number of breeder’s right is ‘5474’

A soybean cultivar for sprout, ‘Joyang 1’ was developed at the National Institute of Crop Science (NICS) in 2010. ‘Joyang 1’ (Iksan67) was released by pedigree selection from the cross between high yielding ‘Pungsannamulkong’ and ‘Suwon187’ tolerant to lodging and disease. It has purple flower, lanceolate leaf, grey pubescence and small yellow seed. Weight of hundred seed is 11.6gram. Maturing date of ‘Joyang 1’ is ten days earlier than check cultivar, ‘Pungsannamulkong’. ‘Joyang 1’ is resistant to bacterial pustule and shattering. It have similar tolerance to root rot and seed spot to check cultivar. It shows high germination ratio, less abnormal germination and hard seed. Sprout yield also 11 percent higher than check cultivar. The average seed yield of ‘Joyang 1’ was 3.03 ton per hectare in the regional yield trials (RYT) carried out for three years from 2008 to 2010 which was 3 percent higher than the check cultivar. The number of breeder’s right is ‘4900’.
