복합내병충성 고품질 중만생 벼 ‘중모1045호’

  • ‘Jungmo1045’, a multi-resistant japonica rice cultivar developed from a cross between ‘Hanggeumnuri’ having a good eating-quality and high yield and ‘SR30071’ having brown planthopper (BPH) resistance, was developed by the rice breeding team of NICS, RDA in 2015. This cultivar has about 121 days growth duration from transplanting to harvesting in west-southern coast, Honam and Youngnam plain of Korea. It has 73cm culm length and 22cm panicle length. In reaction to biotic and abiotic stresses, it shows resistance to bacterial blight pathogen races from K1 to K3, stripe virus and brown planthopper. The milled rice of ‘Jungmo1045’ exhibits translucent, relatively clear non-glutinous endosperm and medium short grain. It has good palatability of cooked rice compared with ‘Nampyeongbyeo’. The milled rice yield performance of this variety is about 5.74 MT/ha in local adaptability test for three years. ‘Junngmo1045’ would be useful genetic resources for multi-resistance breeding program against disease and insect and eco-friendly cultivation.

지구온난화는 전 세계적으로 기후변화의 주요원인으로 알려 져 있다. 한반도의 기온은 지난 100년간 1.7℃가 상승 했다고 보고 됐는데 이는 전 지구의 평균온도 상승률보다 2배 이상 높은 수치이다(NIMR 2009). 이러한 기후변화는 병해충의 발생 을 증가시켜 식량자원의 불균형을 초래할 수 있어 생태적, 경제적 측면에서 광범위한 영향을 미치고 있다(Kwon 2005). 우리나라 도 친환경재배단지의 증가와 더불어 기후변화에 따른 이상고온 으로 벼 병해충의 발생이 늘고 있는데 최근 남서해안가를 중심으 로 벼흰잎마름병과 벼멸구의 피해가 발생하고 있다. 벼흰잎마름 병의 발병 추이는 1970년대 감수성 품종의 재배면적 증가에 따라 발병률이 계속해서 늘어나다 1990년대 관개수로 정비 등으 로 인한 월동기주의 제거로 2000년 초반까지 계속해서 줄어들었 다(Shin et al. 2005). 하지만 2003년 K3 레이스의 변이균 K3a의 출현으로 기존 K3 저항성 품종의 이병화가 급속하게 이루어지면 서 2006년부터 점차적으로 증가, 2010년대에는 매년 벼 재배면 적의 약 1.4% 정도 발생하고 있다(Noh et al. 2003, 2013). 벼멸구 (Nilaparvata Lugens Stảl)의 피해정도와 발생빈도는 1960년대 이후 동남아시아 지역을 중심으로 계속해서 증가하고 있다. 벼멸 구는 동남아와 중국남부지역에서 비래하여 우리나라로 이동하 는데 2013년에는 남서해안가를 중심으로 전체 논의 56%가 벼멸 구 피해를 입었다(National Crop Pest Management System, http://ncpms.rda.go.kr/npms). 병해충의 발생은 농업인 소득과 가장 밀접한 수량 감소에 직접적인 영향을 주는 만큼 저항성 품종을 육성하여 재배하는 것이 가장 경제적이며 자연친화적인 방제 방법이다(Bonman et al. 1992, Kim et al. 2010). 최근 육성된 벼 품종은 기본적으로 병에는 저항성을 가지고 있지만 해충, 특히 벼멸구까지 강한 품종은 아직 많지 않은 실정으로 지금까지 개발된 자포니카형 벼멸구 저항성 벼는 ‘화청’, ‘하남’, ‘다청’, ‘중모1006호’, ‘친농’, ‘안미’, ‘친들’ 7개 품종이다(Kim et al. 2013). 농촌진흥청 국립식량과학원 벼 육종팀은 야생벼 O. australiensis에서 유래한 12번 염색체 장완에 위치하는 Bph18 유전자 연관 분자표지(Jena et al. 2006, Fujita et al. 2013)를 활용하여 자포니카형 벼에 Bph18 유전자와 흰잎마름병 저항성 유전자 Xa3, 그리고 줄무늬잎마름병 저항성 유전자 Stvb-i를 가지는 복합병해충 저항성 계통을 개발하였으며, 그 육성과정을 보고하고자 한다.
시험재료 및 재배방법
‘중모1045호’는 품질이 우수하며 벼멸구 저항성을 갖춘 품종 을 육성하기 위해 모본으로 고품질이지만 벼멸구에 감수성인 중만생 품종 ‘황금누리’를, 부본으로 야생벼 O. australiensis로부 터 유래된 벼멸구 저항성 유전자 Bph18을 가진 벼멸구 저항성 중생종 계통 ‘SR30071-3-7-23-6-2-1-1’을 인공교배 하였다. F2 세대에서는 집단 선발을 하였고 F3 이후부터는 계통육종법으로 선발하였으며 벼멸구 유묘생물검정과 벼흰잎마름병 저항성 생 물검정을 실시하여 저항성이 아닌 계통은 도태하였다. F5 세대에 서는 벼멸구 저항성 유전자 Bph18, 벼흰잎마름병 저항성 유전자 Xa3, 벼줄무늬잎마름병 저항성 유전자 Stvb-i 보유여부를 분자마 커로 판정하였다. F5 세대에서 선발된 계통의 주요 농업형질을 조사하기 위하여 표준품종인 ‘남평’과 함께 2012~2013년 2년간 생산력검정시험을 수행하였다. 재배방법은 재식거리 30×15㎝, 주당 3본씩 이앙 하였고 시험구는 완전임의배치법으로 3반복하 였다. 시비량은 N-P2O5-K2O를 90-45-57kg/ha 시용하였다. 그 밖에 재배관리 및 시비량은 농촌진흥청 표준재배법에 준하였다 (RDA 2012, 2013a, 2014a, 2015a).
주요 병해충 검정
벼멸구 유묘생물검정은 국립식량과학원 해충사육실에서 국 내에서 채집한 벼멸구를 분양 받아 ‘남평’을 먹이로 증식하여 사용하였다. 20칸 육묘판에 상토를 넣고 검정계통종자를 파종하 였고 육묘판의 2개 공간에는 감수성 품종인 ‘남평’과 저항성 품종인 ‘안미’를 함께 파종하여 검정지표로 사용하였다. 접종은 4∼5엽기 정도 되었을 때 2∼3령의 벼멸구 유충 5∼7마리를 개체 당 접종하였으며, 접종 1주일 뒤에 ‘남평’이 완전히 고사한 시점에 저항성 여부를 판정하였다. 벼흰잎마름병 저항성 검정은 우리나라에 분포하는 벼흰잎마름병 4개 레이스의 대표 균주 HB1013(K1), HB1014(K2), HB1015(K3), HB1009(K3a)를 배양하여 사용하였다. 각 균주를 PSA 액체배지에 3일간 배양한 후 분광광도계(Ultraspec 4300 pro, UK)로 병원균의 농도를 108~109cell/㎖로 조정하여 접종하였다. 균 접종은 최고분얼기 인 7월 말에 계통의 엽선단 약 3∼5cm 부위를 가위절엽 접종하 였다. 접종 3주 후 병반길이를 조사 하여 5cm 이하는 저항성, 5∼10cm 중도저항성, 10cm 이상은 감수성으로 판정하였다. 줄무늬잎마름병 저항성 검정은 망실검정법으로 보독매개충을 월동시켜 개체 밀도를 충분히 유지시킨 망실에서 계통종자를 파종한 후 감수성 품종인 ‘일품’이 고사할 때 저항성과 감수성으 로 판정하였다(RDA 2012).
병해충 저항성 유전자 분자표지검정
Genomic-DNA는 계통별 파종 후 2∼3엽기의 어린잎을 1cm 크기로 잘라 마쇄한 후 Biosprint 96(Qiagen) DNA plant kit를 활용하여 추출하였고 Nanodrop(Nanodrop Co.)으로 DNA 농 도를 측정하여 10ng/ul로 조절하여 사용하였다. 본 계통의 벼멸 구, 흰잎마름병 및 줄무늬잎마름병에 저항성 유전자를 탐침하기 위해 각각 7312.T4A, 9643.T4 및 ST10 분자표지를 사용하였다 (Hayano-Saito et al. 2000, Jena et al. 2006). PCR 증폭 조건은 pre-denaturation 95℃ 5분간 1회, denaturation 94℃ 40초, annealing 57℃ 40초, extension 72℃ 1분을 35회, final-extension 72℃ 5분으로 하였다. 벼멸구 저항성 유전자 Bph18과 벼흰잎마름병 저항성 유전자 Xa3 마커 검정을 위해서 는 각각 HinfⅠ(37℃, 2시간 반응)과 TaqⅠ(65℃, 2시간 반응) 제한효소를 처리하고 1.5% agarose gel에서 100V 40분간 전기 영동 한 후 유전자형을 판정하였다.
육성 경위
‘중모1045호’는 품질이 우수하며 벼멸구 저항성을 갖춘 품종 으로 고품질 중만생 ‘황금누리’와 벼멸구 저항성 유전자 Bph18 을 가진 ‘SR30071’ 중생 계통을 인공교배하여 육성하였다(Fig. 1). F1은 40개체를 양성하였는데 그 중 초형과 수형이 우수한 20개체를 선발 하였다. F2는 F1 20개체에서 개체군당 30개체씩 총 600개체를 이앙하였으며 그 중 단간이며 중만생 출수반응을 보이고 이삭과 초형이 우수한 60개체를 선발하였다. 선발한 60 개체 중 쌀외관미질검정을 수행하여 심복백이 있고 투명도 등 외관품질이 안좋은 계통은 도태시키고 최종 23개체를 F3로 공시 하였다. 이후 벼멸구 유묘검정, 도열병 밭못자리검정, 벼흰잎마 름병 K1 균계 접종 등 병해충 특성검정을 실시하여 저항성이며 초형과 수형이 우수한 계통을 선발⋅육성하여 2011년 고정된 우수 F5 계통 ‘HR28011-1-2-3’을 선발하였고 2012∼2013년 2년 간 생산력검정시험을 수행하였다. 수행결과 ‘HR28011-1-2-3’은 내병충성과 함께 기본적인 수량성과 미질을 갖춰 ‘익산562호’로 계통명을 부여하였다. 2013∼2015년 3년간의 지역적응시험에 서 우수성이 인정되어 2015년 12월 직무육성신품종선정위원회 에서 중간모본으로 선정하고 ‘중모1045호’로 명명하였다(Fig. 2, RDA 2013b, 2014b, 2015b).

출수기 및 주요 농업적 특성
‘중모1045호’의 육성은 중만생종 내병충성 품종개발을 목표 로 하였기에 모본과 부본의 생태형은 중만생종이며 ‘중모1045 호’ 또한 출수기는 보통기보비재배 13개 지역에서 3년 평균 8월 14일로 대비품종인 ‘남평’보다 1일 빠른 중만생종이다. 진주에서 가장 빨리 8월 11일 출수하였고 논산과 밀양에서 가장 늦은 8월 16일 출수하였다. 하지만 지대별 평균간에는 통계적으로 유의차는 없었다. 이모작에서도 8월 25일로 ‘남평’보다 1일 빨리 출수하였다. 간장은 78cm로 ‘남평’과 같고 수장은 22cm로 ‘남 평’보다 2cm 길다. 주당수수는 ‘남평’과 같으나 수당립수는 108 개로 ‘남평’보다 13개 많다. 등숙비율은 93.1%로 ‘남평’의 92.6% 와 비교하여 높고 현미천립중은 23.6g으로 ‘남평’보다 1.5g 무거 운 편이다. 수당립수와 현미천립중은 ‘남평’과 비교하여 통계적 으로 유의하였다(Table 1).

생리장해 저항성
‘중모1045호’는 위조와 불시출수는 나타나지 않았으며 성숙 기 하엽노화가 늦은 편이다. 내냉성 검정 결과 출수지연은 ‘남평’ 보다 2일 늦은 10일 이었으며 임실율은 36%로 ‘남평’과 같은 수준이었다. 저온발아성은 85%로 ‘남평’의 37%와 비교하여 매우 높았으며 이와 상관관계가 높은 수발아율도 34.5%로 ‘남 평’의 7%보다 높았고 통계적으로도 유의성을 나타냈다. ‘중모 1045호’의 모본인 ‘황금누리’의 지적시험 수발아율은 6%로 낮 은 편이기 때문에 부본인 ‘SR30071-3-7-23-6-1-1-1’의 수발아 특성을 이어받은 것으로 추측된다(Table 2). 도복 특성은 벼키 (간장+수장)가 102cm, 3절간장이 11cm로 ‘남평’보다 각각 4cm, 2.5cm 길고 좌절중도 1,166g으로 ‘남평’의 1,179g보다 약간 높았다. 도복지수 역시 136으로 ‘남평’보다 높았다. 따라서 도복은 ‘남평’보다 약간 더 잘되는 경향을 가진다고 판단하였다 (Table 3).

병해충 저항성
2013∼2015년 전국 12개소에서 수행한 잎도열병 밭못자리 검정에서 강 2개소, 중 10개소로 ‘남평’과 비슷한 중도 저항성 반응을 보였다. ‘나주’에서는 강한 저항성을 보였는데 전체적으 로 검정포장의 발병상태가 약했던 것으로 추정된다. 목도열병 포장검정에서는 경미한 발병을 보여 목도열병에 저항성을 갖춘 것으로 판단되었다(Table 4). 도열병 내구저항성 검정에서는 40개의 접종균주에서 17개가 친화성균주였으며 내구성 정도는 잎도열병 밭못자리검정과 마찬가지로 중간이었다(Table 5). 벼 흰잎마름병은 K1, K2, K3 레이스에 강하였으나 K3a 레이스에 는 약한 반응을 보였다. 이러한 결과는 모본인 ‘황금누리’의 벼흰잎마름병 저항성 유전자 Xa3를 이어받아 생성된 특성으로 판단된다. Xa3 유전자는 K1, K2, K3 레이스에 저항성을 가지고 있다고 밝혀져 있으며 ‘중모1045호’는 분자표지 검정 결과 Xa3 유전자를 보유하고 있는 것으로 나타났다. 바이러스병 중 벼줄무 늬잎마름병에도 강한 저항성을 가지고 있는데 이는 모본인 ‘황금 누리’가 가지고 있는 저항성 특성을 이어 받은 것으로 판단된다. 하지만 기타 바이러스병인 오갈병과 검은줄오갈병에는 감수성 반응을 보였다. ‘중모1045호’는 벼멸구 유묘검정에서 저항성 반응을 보였다(Fig. 3). 부본인 ‘SR30071’은 O. australiensis로 부터 유래한 Bph18 유전자를 가진 계통으로 자포니카인 ‘주남’ 과 인디카인 ‘IR65482’를 교배하여 육성하였는데 ‘중모1045호’ 는 육성목적에 맞게 ‘SR30071’ 계통이 가지고 있는 특성을 정확 하게 이어받은 것으로 판단된다(Table 6). 또한 벼멸구 저항성 유전자를 분자표지로 확인한 결과 Bph18 저항성 유전자를 보유 하는 것으로 나타났다(Fig. 4).

미질 및 도정특성
‘중모1045호’의 입형은 현미장폭비가 1.69로 ‘남평’의 1.73 과 비슷한 단원형이며 현미길이(장)는 4.86으로 ‘남평’과 통계적 으로 유의하였다. 부본인 ‘SR30071’은 자포니카와 인디카의 교배로 육성된 계통으로 인디카의 특징인 장립 특성이 억제 또는 제거되어 ‘중모1045호’에도 ‘남평’보다 짧은 현미길이를 나타냈다. ‘중모1045호’의 미질은 심복백이 없으며 쌀알이 맑고 투명하였고, 알카리붕괴도와 단백질 함량은 ‘남평’보다 낮으며 아밀로스 함량은 약간 높았다. 밥맛 관능검정 결과 ‘남평’보다 좋은 밥맛을 나타냈지만 평균 밥맛 보다는 못한 평가를 나타냈다. 우리나라 벼 품종 중 Xa3와 Stv-bi 유전자를 가진 품종은 매우 많으며 밥맛이 매우 좋은 최고품질 품종(삼광, 호품, 진수미, 영호진미, 수광, 대보, 현품, 해품, 해담쌀, 청품 등)도 이들 유전자 를 가지고 있다. 하지만 Bph18 유전자를 가진 품종은 2017년 현재 ‘안미’와 ‘중모1045호’ 두 품종 뿐이며 ‘안미’의 밥맛 관능 평가는 ‘추청’ 수준의 밥맛으로 평가되었다. ‘중모1045호’의 모본 황금누리는 고품질 품종으로 밥맛이 중상 이상으로 평가 받고 있기 때문에 현재로서는 부본인 ‘SR30071’의 밥맛 특성을 가지고 있다고 추정 할 수밖에 없다. 하지만 Bph18 유전자 부위가 밥맛에 영향을 미치는지에 대해서는 식미관련 특성이 양적형질 이기 때문에 보다 많은 품종이 육성되어 평가가 이루어져야 할 것으로 생각된다(Table 7). 도정율은 76.1%로 ‘남평’의 73.5% 보다 높았으나 백미완전립률은 93.3%로 ‘남평’의 96.5% 보다 낮은데 백미완전립률은 밥맛에 영향을 주는 요인이므로 불완전 립률을 낮추는 재배법이 필요할 것으로 생각된다(Table 8).

수량성
2013∼2015년 3년간 실시한 지역적응시험 보통기보비재배 13개소에서 5.7MT/ha으로 ‘남평’의 수량과 동일하였다. 지대별 로 영남평야(밀양, 대구, 선산, 진주)에서 6.0MT/ha로 가장 높았 고 서남부해안(계화, 영암, 고성)에서는 5.5MT/ha로 가장 낮은 수량을 보였다. 호남평야(식량원, 예산, 논산, 익산, 나주)에서는 지역간 수량차이가 227kg/10a로 가장 컸다. 이모작재배에서는 2014∼2015년 2년간 2개소에서 5.6MT/ha으로 ‘남평’보다 1% 증수 되었다(Table 9).

적응지역 및 재배상 유의점
‘중모1045호’는 중만생종으로 서남부해안지와 충남 이남 내 륙평야지(충남, 전남북, 경남)에 적합한 품종이다. 키다리병에 약하므로 파종 전 종자소독을 실시하여야 하며 질소질 비료 과용시 미질 및 등숙저하, 숙색불량, 병해충 발생이 우려되므로 적정 균형시비 하여야 한다. 벼멸구, 벼흰잎마름병(K1, K2, K3), 벼줄무늬잎마름병에는 강하나 기타 바이러스병(오갈병, 검은줄 오갈병)에는 약하므로 종자소독 및 적기 방제를 실시하여야 한 다. 수발아율이 높아 등숙기에 비가 지속 될 시 수발아가 우려되 므로 적기수확 하여야 하며 냉해에 약하므로 출수지연이 우려되 는 냉수 용출답 재배는 피해야 한다.
‘중모1045호’는 농촌진흥청 국립식량과학원에서 2015년 고 품질 품종인 ‘황금누리’와 벼멸구 저항성을 가진 ‘SR30071’를 교배하여 육성한 친환경재배적응 고품질 복합내병충성 벼 품종 이다. ‘중모1045호’는 서남부해안지 및 충남이남 평야지(충남, 전남북, 경남) 보통기보비재배에서 이앙부터 수확까지 121일의 생육일수를 가지며 간장은 78cm고 수장은 22cm이다. 병해충 저항성은 벼멸구, 벼흰잎마름병 K1, K2, K3 레이스, 벼줄무늬잎 마름병에 강하지만 기타 바이러스병 및 해충에 약하다. ‘중모 1045호의 쌀알은 맑고 투명하며 종자형태는 중단립종이다. 쌀수 량은 5.7MT/ha로 ‘남평’과 비슷하며 밥맛은 ‘남평’보다 좋은 수준이다. ‘중모1045호’는 복합내병충성 벼 품종으로 친환경재 배단지에 적합하여 안전한 먹거리 생산에 매우 유용한 유전자원 으로 사용될 수 있을 것으로 생각된다.
