  • Fusarium wilt is a serious disease that causes damping-off, serious wilt symptoms, or death in melon. The aim of this study was to evaluate the resistance of melon germplasm to Fusarium wilt and select promising disease-resistant accessions for further applications. Resistance to Fusarium wilt was examined in 216 melon germplasms by the root-dipping inoculation method with Fusarium oxysporum f. sp. melonis fungal isolate KACC43206 (race 2). Inoculated seedlings were incubated for four weeks and assessed for disease symptoms using indices ranging from index 1 (no symptoms) to 5 (plant wither). A total of 34 accessions showed a disease index < 2.0. Disease indices of 10 accessions were 1.0 four weeks after inoculation. The resistant accessions of melon are expected to help in breeding commercial varieties with Fusarium wilt resistance.
멜론 덩굴쪼김병은 1930년 미국 뉴욕에서 처음 보고(Chupp 1930)된 이래 미국(Leach & Currence 1938)뿐만 아니라 유럽, 아시아, 아프리카 등(Schreuder et al. 2000, Sherf & Macnab 1986, Quiot et al. 1979) 전 세계 적으로 발생하여 문제가 되고 있다(Appel & Gordon 1994, Katan et al. 1994, Zitter 1999).
멜론 덩굴쪼김병균(Fusarium oxysporum f. sp. melonis)은 토양 전염성 균으로 후막포자를 생성하여 장기 생존하며(McKeen & Wensley 1961), 식물의 뿌리를 통해 침입하여 도관에서 증식한다(Bishop & Cooper 1983, Zvirin et al. 2010). 이로 인해 식물은 시들고 심하면 고사하게 된다(Sherf & Macnab 1986). 멜론 덩굴쪼김병균(F. oxysporum f. sp. melonis)은 race 0, 1, 2, 1,2 4개의 race가 있는 것으로 알려져 있으며(Risser et al. 1976), 미국(Zuniga et al. 1997) 유럽(Katan et al. 1994, Elena & Pappas 2006), 일본(Namiki et al. 1998, 2000)에서 race의 분포에 대한 보고가 이루어졌다.
멜론 덩굴쪼김병에 대한 저항성 유전자는 Fom-1 (Messiaen et al. 1962)과 Fom-2 (Risser et al. 1976)가 보고되어 있으며 그 후로도 저항성 자원을 찾기 위한 노력이 계속되었다(Zink et al. 1983, Champaco et al. 1992, Pitrat et al. 1996, Alvarez et al. 2005).
멜론 덩굴쪼김병 저항성 유전자 Fom-1Fom-2을 바탕으로 분자표지가 개발되어 육종에 활용되고 있으며(Kim 2020, Oumouloud et al. 2013), 미국, 일본, 프랑스 등에서는 race 0, 1, 2에 저항성인 멜론 품종이 판매되고 있다(Matsumoto et al. 2011, Zink & Thomas 1990). 그러나 우리나라에서는 멜론 덩굴쪼김병균의 race가 알려져 있지 않으며, Lee et al. (2018)이 국내에 판매되고 있는 멜론 22품종의 저항성을 조사한 결과, race 2에 저항성을 보이는 품종이 없었다. 따라서 본 연구에서는 저항성 연구나 육종에 활용 가능한 멜론 덩굴쪼김병 race 2에 저항성인 멜론 유전자원을 탐색하고자 농업유전자원센터에서 분양 받은 멜론 유전자원 216자원을 대상으로 멜론덩굴쪼김병균 race 2에 대한 저항성을 평가하였다.
실험재료
농촌진흥청 국립농업과학원 농업유전자원센터에서 멜론(Cucumis melo) 유전자원 216자원을 분양 받아 덩굴쪼김병 저항성 검정에 이용하였다. 대조품종으로는 얼스베타리치, 얼스마운틴, 아시아황금, 얼룩파파야를 이용하였다. 종자는 물에 4시간 침지 후 48시간 동안 31℃에 건조하였다. 침지 후 건조한 종자를 32구 트레이에 자원당 8립씩 파종하여 온실에서 육묘하였다.
병원균 배양
국립농업과학원 미생물은행(KACC)에서 F. oxysporum f. sp. melonis KACC43206 균주를 분양 받아 실험에 이용하였다. KACC43206균주는 Lee et al. (2018)가 멜론 덩굴쪼김병 판별 품종(Charentais T, Charentais Fom-1, Charentais Fom-2, MR-1)을 이용하여 race를 동정한 결과, race2로 나타났다. 분양 받은 KACC43206 균주를 Potato Dextrose Agar (PDA)배지에 접종하여 7일간 25℃에서 배양한 후, 균이 배양된 PDA배지를 직경 6 mm로 조각내어 V8 액체배지(V8 juice 70 mL, 증류수 630 mL)에 10조각을 접종하여 4일간 25℃, 암조건에서 진탕 배양기를 이용하여 150 rpm으로 진탕 배양하였다.
접종 방법
유전자원의 덩굴쪼김병 저항성을 검정하기 위해 뿌리침지 법을 이용하였다. 1~2엽기(파종 후 14일경)의 유묘를 뽑아 1×106 conidia/mL 농도의 균주 현탁액 200 mL에 20분간 담근 후 32구 트레이에 다시 심었다. 접종한 유묘는 2일간 덮개를 씌워 습도를 유지해 주었으며, 접종 후 4주간 25℃에서 재배하였다.
발병도 조사
접종 4주 후 발병도와 발병율을 조사하였다. 발병도 조사기준은 1에서 5까지 무발병은 1, 지상부 생육은 억제되지 않았으나 지하부는 갈변된 것은 2, 지하부가 갈변되고 지상부 생육이 억제된 것은 3, 지하부가 갈변되고 지하부, 지상부 생육이 상당히 억제된 것은 4, 고사는 5로 조사 하였다(Fig. 1). 자원당 1반복 당 8개체씩 2반복 실험하였고, 발병도의 평균을 계산하여 평균±표준오차로 표기하였다. 자원의 평균 발병도가 2.0이하는 저항성, 2~4는 중도저항성, 4이상은 감수성으로 판단하였다.
국립농업과학원 농업유전자원센터로부터 분양 받은 멜론 유전자원 216자원에 멜론 덩굴쪼김병균(F. oxysporum f. sp. melonis) KACC43206을 뿌리침지법을 이용하여 접종한 결과, 접종 4주 후 자원들의 발병도는 1.0에서 5.0까지 분포하였으며, 평균 발병도는 3.6이었다. 대조자원은 4자원 모두 발병도 4.5이상으로 나타났으며, 국내시판 멜론품종의 덩굴쪼김병 저항성을 조사한 Lee et al. (2018)의 논문에서도 본 논문에서 사용한 대조품종 4자원 모두 race 2 균주인 KACC43206에 대해 감수성을 나타내었다.
216자원을 발병도 정도 별로 나누어 보면 발병도 1.0이 10자원, 발병도 1이상 2미만은 24자원, 발병도 2이상 3미만이 10자원, 발병도 3이상 4미만은 9자원, 발병도 4이상 5미만이 55자원, 발병도 5인 자원이 108자원이었다(Fig. 2). 실험결과 저항성(발병도 2이하)가 34자원(15.7%), 감수성(발병도 4이상)이 163자원(75.5%)인 반면 중도저항성(발병도 2~4)은 19자원(8.8%)로 나타나 중도저항성 자원의 비율이 낮고 저항성 혹은 감수성으로 뚜렷하게 나뉘는 양상을 보였다(Fig. 3). 본 실험에 사용한 F. oxysporum f. sp. melonis KACC43206은 Lee et al. (2018)이 race 2로 동정하였는데, 멜론 덩굴쪼김병 저항성 유전자는 race 2에 저항성인 Fom-1와 race 1에 저항성인 유전자 Fom-2가 있으며(Risser et al. 1976) 둘 모두 단인자 우성유전을 한다고 알려져 있어(Risser et al. 1976, Messiaen et al. 1962) 본 실험의 결과와 일치하였다.
멜론 216자원의 원산지는 터키 47자원, 중국 45자원, 한국 35자원, 우즈베키스탄 32자원 등 19개국이었고, 그 중 중국 자원의 경우 발병도 2 미만인 자원이 45자원 중 22자원으로 저항성을 보이는 자원이 많았다(Table 1). 자원구분 별로는 재래종 139자원, 육성품종 42자원, 육성계통 31자원 등이었고, 육성품종에서 발병도 2 미만인 자원의 비율이 높았다(Table 2).
발병도 2 미만인 34자원의 원산지는 중국 22자원, 한국과 우즈베키스탄 3자원씩, 터키 2자원, 러시아, 인도, 불가리아, 일본이 1자원씩이었다(Tables 1, 3). 자원 구분 별로는 육성품종(16자원), 재래종(14자원), 육성계통(3자원), 잡초형(1자원) 순이었다(Tables 2, 3). 34자원 중 발병도 1.0, 발병주율 0%로 나타나 강한 저항성을 보인 10자원은 중국 육성품종 4자원, 중국 재래종 2자원, 그리고 한국 잡초형, 러시아 재래종, 우즈베키스탄 육성계통, 일본 육성품종이 1자원씩이었다(Table 3).
Messiaen et al. (1962)은 프랑스의 캔탈로프 멜론 품종들 중 멜론 덩굴쪼김병에 저항성인 ‘Doublon’과 ‘Vedrantais’를 선발했으며, 이들 품종의 저항성이 race 0과 2에 저항성인 유전자 Fom-1에 의한 것임을 밝혀냈다. 또한 Risser et al. (1976)은 ‘CM17187’에서 race 0과 1에 저항성인 유전자 Fom-2을 보고하였다. 이후에도 Zink et al. (1983)이 멜론 152자원의 멜론 덩굴쪼김병 race 2에 대한 저항성을 조사하여 32자원이 저항성을 나타내었고, Pitrat et al. (1996)은 353자원 중 52자원이 race 0과 2에, 49자원이 race 0과 1에 저항성을 나타내었다고 하였다. Alvarez et al. (2005)은 멜론 139자원의 덩굴쪼김병균 race 0, 1, 2 세 race에 대한 저항성을 평가한 결과 race 1에 저항성인 자원이 4자원, race 0과 2에 저항성인 자원이 12자원으로 나타나 race 1에 저항성인 자원이 적었다고 하였다.
미국(Zuniga et al. 1997) 유럽(Katan et al. 1994, Elena & Pappas 2006), 일본(Namiki et al. 1998, 2000)에서는 멜론 덩굴쪼김병균(F. oxysporum f. sp. melonis)의 race의 분포에 대한 보고가 이루어졌으며, race 0, 1, 2에 저항성인 멜론 품종이 판매되고 있다(Matsumoto et al. 2011, Zink & Thomas 1990). 그러나 우리나라에서는 멜론 덩굴쪼김병균의 race가 알려져 있지 않으며, Lee et al. (2018)이 국내에 판매되고 있는 멜론 22품종의 저항성을 조사한 결과, 19개품종이 race 0과 1에 저항성을 보였으나, race 2에 저항성을 보이는 품종은 없었다. 이를 고려할 때 본 연구가 유묘기에만 저항성 검정이 이루어졌음을 고려할 필요가 있기는 하지만, 본 연구에서 저항성을 보인 자원들은 향후 저항성 품종 육종 재료나 연구 소재로 활용 될 수 있을 것으로 기대된다.
멜론 덩굴쪼김병을 일으키는 F. oxysporum f. sp. melonis race 2에 저항성인 멜론 유전자원을 탐색하기 위하여 농촌진흥청 국립농업과학원 농업유전자원센터로부터 멜론 216자원을 분양 받아 뿌리침지법을 이용하여 접종한 뒤 4주후 발병도를 1~5로 조사하였다. 그 결과, 216자원의 발병도는 1.0에서 5.0까지 분포하였으며 평균 발병도는 3.6이었다. 216자원중 발병도 2.0이하인 자원이 34자원이었으며, 그 중 10자원은 발병도가 1.0으로 나타났다. 본 연구에서 저항성을 보인 자원들은 저항성 연구와 품종 육성에 활용할 수 있을 것으로 기대된다.
